Extraction and purification of Debregeasia orientalis fruit polyphenols and its antioxidant,antitumor activity

Abstract: In order to extract polyphenols from Debregeasia orientalis fruit and measure it's antioxidant and anti-tumor activity,the polyphenol extraction technology was optimized by single factor experiment and orthogonal experiment.Further,the Debregeasia orientalis fruit polyphenols were purified by macroporous resin.The antioxidant capacity of Debregeasia orientalis fruit polyphenols was characterized by measuring its total reducing power and the scavenging capacity of ·OH,DPPH· and ABTS·.Human cervical cancer Hela cells and human non-small lung cancer A549 cells were used as the anti-tumor object to measure its anti-tumor ability.The results indicated that the highest yield of Debregeasia orientalis fruit polyphenols assisted by ultrasonic extraction could reach 3.29% while under the condition 60% ethanol,70 °C and 40 min extraction,and the total polyphenol content of purified product was 40.47 mg/100 mg.The results of in vitro antioxidant tests showed that the total reducing power of Debregeasia orientalis fruit polyphenols was similar to that of vitamin C and had significant scavenging effect on ·OH,DPPH· and ABTS·.Besides,Debregeasia orientalis fruit polyphenols had a significant proliferation-inhibiting effect on Hela cells and A549 cells,and led to lots of reactive oxygen species(ROS) produced in cancer cells,combined with the morphological features of apoptosis.Overall,the total polyphenol content of the extract is significantly increased after purification by the macroporous resin,and has significant anti-oxidation and anti-tumor activity.
