Antiglycation activity,acetylcholine esterase and α-glucosidase inhibitory activity of total flavonoids from corn silk

Abstract: To study the antiglycation activity,acetylcholine esterase and α-glucosidase inhibitory activity of total flavonoids from corn silk.Seven parameters potentially influencing the extraction yield of total flavonoids were screened through Plackett-Burman design to determine the significant variables.Then,factors effecting on extraction yield were further optimized using Box-Behnken design to predict optimum extraction conditions.Satisfactory yield of total flavonoids (4.75±0.69%)was achieved using the optimal conditions.16 min of desorption time,73% of ethanol indesorption solvent,20 min of extraction time,and 61 ℃ of extraction temperature were selected and applied for effective extraction.It also exhibited the certain antiglycation,inhibitory activity against acetylcholine esterase and α-glucosidase in a concentration-dependent manner with IC50 of 467.90,73.07,and 151.04 μg/mL,respectively.Antiglycation activity,acetylcholine esterase and α-glucosidase activity inhibitory activity were positively correlated with the content of total flavonoids,whose correlation coefficients are 0.83,0.90 and 0.87 respectively,P<0.05.It can provide a theoretical basis for the development of natural inhibitors.
