A new daphnane diterpenoid from the flower buds of Daphne genkwa

Abstract:

To study the chemical constituents and their anti-dermatophyte activities of Daphne genkwa.Two daphnane diterpenoid derivatives were isolated from the 95% ethanol extract of Daphne genkwa using various column chromatography,such as silica gel and preparative HPLC.And the structures were identified by NMR,MS,and ECD spectroscopic techniques,as well as comparison with literatures.Compound 1 was a new daphnane diterpenoid compound,and named as daphgenin A (1),compound 2 was identified as yuanhuakine B (2).The anti- dermatophyte activities of the obtained compounds against Microsporum canis,Trichophyton rubrum,Trichophyton mentagrophytes and Malassezia restricta were evaluated by MIC method.The results showed that compounds 1 and 2 displayed certain inhibitory activity against dermatogenic fungi.The inhibitory activities of 1 against T. rubrum and T. mentagrophytes with MIC value 4.0 μg/mL,were equivalent to that of the positive control group,and compound 2 showed significant inhibitory activity against T. rubrum with MIC value 2.0 μg/mL.
