Medicinal value of Lablab Semen Album and its seed coat based on HPLC fingerprint and chikusetsusaponin IVa content analysis

Abstract:

This study aimed to establish the HPLC fingerprint profiles of Lablab Semen Album (LSA) and its seed coat (LSAC),and the content determination method for chikusetsusaponin IVa (CS-IVa),and compare them with Dolichos lablab seed with black seed coat (black lentils) and with red seed coat (red lentils),as well as 17 other types of beans to explore their medicinal value.HPLC was used to establish the characteristic profiles,and chemometric methods including hierarchical cluster analysis (HCA),principal component analysis (PCA),orthogonal partial least squares discriminant analysis (OPLS-DA) were applied to perform chemical pattern recognition analysis on different types and parts of lentils and other beans.The results showed that the HPLC fingerprint profiles of LSA and LSAC were established with 14 common peaks identified,including two common chromatic peaks (protocatechuic acid and soyasaponin) and two differential characteristic peaks (genistin and CS-IVa).HCA classified 15 batches of samples into two categories,and OPLS-DA analysis screened out seven common characteristic peaks affecting quality.CS-IVa was determined as the quantitative analysis indicator and was mainly concentrated in the seed coat.The study results indicated that LSA and LSAC were clearly distinguished from other common beans in terms of HPLC fingerprint profiles and CS-IVa content,providing a scientific basis for elucidating their medicinal value.
