Identification and Purification of Paeoniflorins in Seed Cakes of Paeonia sufruticosa by Macroporous Adsorption Resin

Abstract: Four main paeoniflorins,namely 6′-O-β-D-glucopyranosylalbiflorin,albiflorin, β-gentiobiosylpaeoniflorin and paeoniflorin were isolated from the alcohol extract of seed cakes of Paeonia sufruticosa Andr.and identified by LC-MS and1H-NMR.The purification procedure using macroporous adsorption resins was optimized.The results showed that HPD200A resin was the most suitable resin among the 11 tested resins.The optimal procedure for the purification was as follows:the concentration of sample solution was 8.0 mg (paeoniflorin)/mL,the loading capacity was 4.5 bed volume (BV) and the flow rate was 1/16 BV/min;then eluted with 50% ethanol for 4 BV with elution flow rate of 1/16 BV/min.Under these optimal conditions,the content of paeoniflorin,albiflorin,6′-O-β-D-glucopyranosylalbiflorin and β-gentiobiosylpaeoniflorin was reached to 32.3%,16.5%,8.02% and 6.63%,respectively.
