Optimization of microwave-assisted cellulase extraction of polyphenols from Moringa oleifera seeds and its activity

Abstract:

To optimize the extraction process of polyphenols from Moringa oleifera seeds,microwave-assisted cellulase method was used for extraction.The effects of enzyme addition amount,microwave time,enzymatic hydrolysis time,and ethanol volume fraction on the extraction amount of polyphenols from M.oleifera seeds were studied,and response surface methodology was used to optimize the extraction process.The results showed that the optimal conditions were:enzyme addition of 0.4%,ethanol volume fraction of 55%,enzymatic hydrolysis time of 35 min,microwave time of 7 min.The extraction amount was 19.23 mg/g,which was not significantly different from the predicted value and was 3 to 5 times higher than the extraction amount obtained by traditional methods.When the concentration of polyphenols from M.oleifera seeds was 10.24 mg/mL,the clearance rates of ABTS and DPPH free radicals reached 91.4% and 90.7%,at the same concentration,the clearance efficiency respectively reached 91.6% and 90.8% of VC,and the inhibition rate of α-amylase reached 66.8%,indicating that polyphenol from M.oleifera seeds extract has good antioxidant activity and inhibitory ability on amylase.
