    Baicalin Improves DSS-Induced Colitis in Mice Via the TLRs/MyD88 Signaling Pathway

    The objective of this study was to elucidate whether baicalin reduces colonic inflammation in a mice colitis model through inhibition of the TLRs/MyD88 signaling pathway.C57BL/6 mice were randomized into control group,DSS group (administered with 3.5% DSS solution) and baicalin group (administered with 3.5% DSS solution and orally administered with baicalin,30 mg/kg body weight).The disease activity index (DAI) score and histological score were evaluated.Myeloperoxidase activity was detected and colonic mucosal histological changes were observed by HE staining.The protein expression levels of Tumornecrosis factor-α (TNF-α) and interleukin-6 (IL-6) levels in colonic tissue were determined by ELISA.The mRNA expression levels of TLR2,TLR4 and MyD88 in colonic tissue were detected by Real-time PCR.Compared with the DSS group,the baicalin-treated group showed significant decreases in DAI,histological score,myeloperoxidase activity, expressions levels of TNF-α protein,IL-6 protein,TLR2 mRNA,TLR4 mRNA and MyD88 mRNA.Collectively,these findings highlighted that baicalin had significant therapeutic effects on DSS-induced colitis and the anti-inflammatory actions of baicalin may involve inhibition of the TLRs/MyD88 signaling pathway.

    Extraction of Total Flavonoids from Scutellariaby Cellulose Assisted Aqueous Ethanol

    In order to optimize the extraction process of Scutellariaand increase the extractionyield of total flavonoids,ethanol reflux extraction,cellulose-aqueous buffer solution extraction and cellulose assisted aqueous ethanolextractionwere compared using the yield of total flavonoids as index.The results showed that there was a synergistic effect between enzymatic hydrolysis of cell walls and the flavonoidsleaching inethanol aqueoussolution.The optimal conditions of cellulose assisted aqueous ethanolextractionwasas follows:extraction temperature 50 ℃,ethanol 50%(v/v),liquid to solid ratio 15,extraction time 6 h,cellulose dosage 50 U/g (dry ingredients).Under the optimal conditions,the extractionyieldof the flavonoidswas19.85%,1.38% higher than the extraction yieldwithoutenzymatic hydrolysis.Hence,it was concluded that cellulose assisted aqueous ethanolextraction method can be used for the extraction of the total flavonoids from Scutellaria.

    Ionic Liquid-based Microwave-Assisted Extraction of Four Flavonoids from Scutellariae Radix Slices

    A rapid reversedphase high performance liquid chromatography method was developed for the simultaneous determination of 4 flavonoids,namely baicalin,wogonoside,baicalein and wogonin.The chromatographic separation was performed on an Agilent TC-C18 column(250×4.6 mm,5.0 μm)with 0.4% phosphoric acid(A)and methanol(B)as mobile phases.The gradient elution program was as follows:0-10 min,40%-50%B;10-20 min,50%B;20-30 min,50%-60%B;30-40min,60%-80% B;40-50 min,80%B.The detection wavelength was set at 280 nm.Under the developed HPLC method,the ionic liquid-based microwaveassisted extraction(IL-MAE)of 4 flavonoids from Scutellariae Radix slices was optimized.The optimal IL-MAE conditions were determined as follows:1.0 M 1-butyl-3-Methylimidazolium bromide([C4 min]Br)employed as extractant,the ratio of solid to liquid was 1:100,extracting for 3 min,the microwave power was 264 W.Compared with traditional solvent reflux extraction(SRE)recorded in Chinese pharmacopoeia(2010 edition),IL-MAE significantly improved the extraction yields of 4 flavonoids and shortened the extraction duration(from 3 h to 3 min).In conclusion,the developed HPLC method was sensitive,accurate,specific,and applicable for the quality control of Scutellariae Radix slices,and the optimized IL-MAE method was a simple,efficient,rapid,and green extraction technique.

    The paper established a method for separation and determination of total flavonoids and three main components in Scutellaria Radix total flavone aglycone extract by ultraviolet (UV) spectrophotometry and high-performance liquid chromatography.The anlysis was performed on an Agilent 1200 Series HPLC system with Eclipse XDB-C18 (250 mm×4.6 mm,5 μm) column.The mobile phase consisted of methanol-0.2% phosphoric acid water (50:50) with the flow rate of 1 min/mL.The column remained at 35 ℃ and the detection wavelength was set at 275 nm.The linear ranges of baicalein,wogonin and oroxylin A were 3.501-70.016 μg/mL (r=0.9999), 0.832-16.640 μg/mL (r=0.9998) and 0.418-8.352 μg/mL (r=0.9999),respectively.The average recoveries (n=6) were 99.55% (RSD=1.74%),98.86% (RSD=1.74%) and 99.97% (RSD=1.70%).Total flavonoids were determined by UV spectrometry.The linear range of baicalein was 1.71-8.54 μg/mL (r=0.9991).The average recoveries (n=6) were 100.93% (RSD=2.63%).The contents of baicalein,wogonin,oroxylin A and total flavonoids were 48.22%-60.36%,12.09%-14.46%, 5.05%-8.81% and 78.11%-90.12%,respectively.

    Optimization of Extraction of 3 Traditional Chinese Medicines with Antibacterial Activity

    In order to establish the optimal methods for the extraction of 3 traditional Chinese medicines,the extractive yield,minimum inhibition concentration (MIC) and minimal bactericidal concentration (MBC) against Streptococcus agalactiae were used as comprehensive evaluation indexes.L9(34) orthogonal design was applied to select the optimal extraction conditions of 3 traditional Chinese medicines.The results showed that the optimal extraction conditions of 3 traditional Chinese medicines were as follows:Coptidis Rhizoma was added to 8 times of 80% ethanol,which were refluxed and extracted for 2 times with each time of 1.5 h;Radix Scutellariae were extracted with 10 times of 55% ethanol at 95 ℃,and repeated for 3 times,2 h for each time; Polygonum cuspidatum extracted 3 times at 95 ℃ with 5 times of 60% ethanol,2 h for each time.Under optimal conditions,the MICs of the extracts obtained from Coptidis Rhizoma,Radix Scutellariae and Polygonum cuspidatum against  S.agalactiae  were 26.04,5.20 mg/mL and 15.60 mg/mL,respectively and the MBCs were 26.04,6.50,31.25 mg/mL respectively.The developed extraction method was simple,rapid,accurate,reproducible and stable.

    In this study,the content of main effective ingredients including baicalin,baicalein and wogonin of wild Scutellaria baicalensis from 10 areas of Shanxi were investigated by HPLC.The results showed that:there was a significant difference in the content of baicalin,baicalein and wogonin in 10 samples of Scutellaria.The content of baicalin,baicalein and wogonin were 5.79%-15.31%,0.49%-2.28% and 0.07%-0.42%,respectively.The content of baicalin had positive correlation with the cultivation latitude of S. baicalensis,the content of baicalin in S. baicalensis samples from Goubei country,Youyu county and Yinliangshan,Shanyin county was not match up with the standard (8.0%) required by Chinese Pharmacopoeia,while the content of baicalin from Wayaoshang country,Linchuan county was determined to be 15.31%.The content of baicalein and wogonin from Suangjiazhai country,Jiaocheng county was determined to be 2.28% and 0.42%.

    Inhibitory Mechanism of Baicalin on H5N1 Influenza Virus

    To study the inhibitory activity and mechanism of baicalin against H5N1 pseudovirus.Inhibition of viral-entry by baicalin was determined with H5N1 pseudoviruses system infected with different H5N1 pseudovirus strains,while VSVG pseudoviruses was used as negative control.Hemagglutination inhibition assay and enzyme-linked immunosorbent assay (ELISA) were performed to investigate the mechanism for its anti-influenza activity.The cytotoxicity of baicalin was measured using MTT assay.The results demonstrated that baicalin specifically inhibited the entry of H5N1 pseudovirus strains,including A/Anhui/1/2005,A/Xinjiang/1/2006,A/Hong Kong/156/1997,A/Qinghai/59/2005,A/Thailand/Kan353/2004,A/VietNam/1194/2004,and the IC50 values were 65.76±5.61 μM,54.98±4.38 μM,46.81±5.12 μM,32.88±4.18 μM,63.30±1.59 μM and 43.23±3.43 μM,respectively.Moreover,baicalin targeted HA2 subunit of hemagglutinin with IC50 of 35.86±3.09 μM.These findings suggested that baicalin blocked entry of H5N1 influenza virus directly.The action mechanisms may be potential target to HA2 submit of hemagglutinin.

    The aim of this study was to investigate protective effect of baicalin on LPS induced rabbit mastitis.Fifty healthy rabbits after parturition 7~10 d were divided into five groups randomly,including mastitis model control group (LPS,0.25 mg/kg·BW),baicalin groups with the dose 75,150 and 300 mg/kg·BW respectively,and dexamethasone (DEX) group with dose of 0.5 mg/kg·BW DEX.LPS was respectively infused into the fourth mammary glands of rabbit via the teat duct.In the baicalin and DEX groups,baicalin and DEX were injected through the auricular veins at 48 h,24 h,1 h before LPS infusion and after 8 h,32 h,56 h respectively.The rectal temperature was measured and the auricular veins blood were collected 50 h before,and 6,12,24,48,72 h after LPS infusion respectively.The following indexes at different time point were determined:(1) The pathological change of rabbit mammary gland tissues.(2) The activities of ALP,LDH,myeloperoxidase (MPO) and N-Acetyl-β-D-Glucosaminidase (NAG) in serum.(3) The levels of TNF-α,interleukin-1 (IL-1) and interleukin-6 (IL-6) in serum were measured by ELISA.The results showed that:(1) Baicalin could reduce the histopathological damage.(2) Baicalin could reduce the activities of ALP,LDH,MPO and NAG.Baicalin group (300 mg/kg·BW) had the best effect of decreasing in the activities of ALP,LDH,MPO and NAG.(3) Baicalin could reduce the level of TNF-α,IL-1,and IL-6 in serum.The level of TNF-α in serum was significantly lower than model control group in 6 h (P<0.05).The level of IL-1 in serum was significantly lower than model control group in 72 h (P<0.05).The results demonstrated that baicalin can reduce the histopathological damage and relief body symptom.The highest potent protective effect of baicalin on LPS induced rabbit mastitis was showed at the dose of 300 mg/kg·BW.

    To develop a new method of obtaining Scutellaria Radix total flavone aglycone.The pH of baicalinasewas optimized using enzymatic hydrolysis rate of baicalin as evaluation index.Taking extraction yield of baicalin as evaluation index,parameters such as water volume,decocting time and extraction times were optimized by orthogonal test.The established method:stirring the Scutellariae Radix powder with water (pH3) for 0.5 h,then baicalinase solution was obtained by filtration,the residue was put into boiled water (12 times of residue) two times for decocting at pH6,0.5 h each time,then mixed the filtration fluid with the baicalinase solution at 50 ℃(pH6) for 6 h.Scutellaria Radix total flavone aglycon can be obtained by drying the precipitate of filtration.The extraction rate of baicaleinwas 80%.The developed method was simple,reliable and stable.It can be used as a way to get Scutellaria Radix total flavone aglycon from Scutellaria Radix.

    In this study,the properties of baicalein such as whitening,antioxidant,antiallergy,antibacterial and safety were evaluated using biochemical enzyme method,free radical scavenging assay,determining the minimum inhibitory concentration (MIC) and experimenting hemolysis.The results showed that the inhibition rate of tyrosinase was 60.20% (2.0 mg/mL of baicalein),the scavengingrate of DPPH· and ·OH was 95.21% and 66.69% respectively (0.1 mg/mL and 1.0 mg/mL of baicalein),and the inhibition rate of enzyme was 52.51% (10.0 mg/mL of baicalein).In addition,when the concentration of baicalein reached 1.25 mg/mL,it had inhibitory effect on the five kinds of bacteria.Its half hemolysis rate (HD50) was 1.80 mg/mL,protein denaturation index (DI) was 0.34,and HD50/DI (H/D) >10.The experiments proved that baicalein exhibited better performance in whitening,antioxidant,antiallergy and antibacterial effects.

    In this study,the HPLC fingerprint method of the pieces,standard decoction,intermediates and dispensing granules of Scutellaria baicalensis was established,and the correlations among them were evaluated.The chromatographic separation was carried out on an Agilent Eclipse XDB C18 column (250 mm×4.6 mm,5 μm);The mobile phase was eluted with gradient by methanol-0.05% phosphoric acid;The flow rate was 1.0 mL/min.The column temperature was 35 ℃ and the detection wavelength was set at 275 nm.There were 9 common characteristic peaks in all fingerprint spectrum of 10 batches of pieces,standard decoction,intermediates and dispensing granules of S.baicalensis,presenting a good correlation,in the 9 common characteristic peaks,three of them were baicalin,baicalein,wogonin.The main chemical constituents of S.baicalensis pieces,standard decoction,intermediates and dispensing granules composition are basically identical.The established HPLC fingerprint method can be used for the quality control of S.baicalensis dispensing granules.

    In this study,mid-infrared spectroscopy was used to distinguish Radix scutellariae granule from different manufacturers.The original infrared spectra and derivative spectra of Radix scutellariae granule from six manufacturers were analyzed.The similarity of original spectrum of sample from same manufacturer was very big.The original infrared spectra and the first derivative spectra cannot distinguish different manufacturers,although some peak position,intensity and shape were different.The peak position,intensity,shape of the characteristic absorption peak of the second derivative spectra in 2914-2900,2887,1733,1500,1218-1206,994,763 cm-1 were obvious different.The results showed that using second derivative spectra of sample can successfully identify Radix scutellariae granule of six manufacturers.This method is fast,simple,nondestructive,accurate and green.

    Baicalin is a kind of flavonoids and extracted from the roots of Traditional Chinese Medicine Scutellaria baicalensis Georgi.It has antibacterial,diuresis,anti-inflammatory,spasmolysis and antitumor function.However,baicalin has hygroscopicity due to the polar groups of the hydroxyl and the carboxyl,so the objectives of this study were to investigate the effects of temperature and relative humidity on the hygroscopicity characteristics of baicalin,and to develop an adsorption isotherm model.Results showed that low temperature and high relative humidity led to higher equilibrium moisture content for moisture adsorption processes.At higher temperature,the equilibrium moisture content was reached over a shorter time indicating a higher adsorption rate;relative humidity was also positively correlated with the adsorption rate.To fit experimental data results indicated that the double exponential model was the most suitable for fitting the moisture adsorption kinetics curve and the Peleg model was the most suitable for fitting the adsorption isotherm with R2 = 0.9984 for baicalin by the 1stOpt statistical analysis software package.

    To study the antibacterial effect of combination of the stem and leave of Scutellaria baicalensis (SLSB) and Ampelopsis grossedentata (AG) aginst Staphylococcus aureus and Shigella dysenteriae,the micro broth dilution method was used to detect their MIC values.The time-bactericidal curves of SLSB and AG against S.aureus and S.dysenteriae were established as well.A checkerboard method and the fractional inhibitory concentration index (FIC index) were used to evaluate the interaction between SLSB and AG.The results showed that the MIC values of SLSB and AG against S.aureus were 1 mg/mL and 0.25 mg/mL,while against S.dysenteriae were 4 mg/mL and 0.50 mg/mL,respectively.And the FIC indices of combined SLSB and AG,0.750 for S.aureus and 0.625 for S.dysenteriae,revealed their additive interaction against the tested bacterials.The present experimental results thus indicated the potential to increase the antibacterial efficacy of SLSB by rational combination strategy,providing a practical way for the rational utilization of SLSB resource.

    This study mainly discussed the protective effect of baicalin on AC16 cardiomyocytes injured by Lipopolysaccharide.The experiment was divided into control group,LPS group,baicalin high,medium and low dose groups,total flavonoids of astragalus membranaceus group and cultured for 6 hours to test cell survival rate and survival status;The firefly luciferin method detects changes in intracellular ATP concentration;The expressions of Cyt C,Apaf-1,caspase-3 and caspase-9 in each group were detected by Western blot.The results showed that compared with the LPS group,the cell survival rate and survival status of the high,middle and low doses group of baicalin and the total flavonoids of astragalus membranaceus group were significantly improved,the intracellular ATP concentration increased,and the apoptotic rate decreased.The expression of Cyt C,Apaf-1,caspase-3 and caspase-9 was decreased in cells.It indicated that baicalin has protective effect on AC16 cardiomyocytes damaged by LPS,and its mechanism may be related to the decrease of Cyt C/Apaf-1/caspase-3/caspase-9 pathway expression.

    Structural modification and antitumor activity of baicalein

    In order to improve the anti-tumor activity of baicalein,it was structurally modified.First,an amine methylene group is introduced to the 8-position by a mannich reaction,and then a different hydrophobic group is introduced to the 7-position (6-position) phenolic hydroxyl group by an acylation reaction.The target compound was evaluated for activity against MCF-7 tumor cells by the CCK-8 method.As a result,six target compounds were synthesized,and compound 2-6 are new compounds.Their structures were determined by a combination of 1H NMR,13C NMR,MS and chemical methods.The chemical methods utilized the characteristics of the o-diphenol hydroxyl group of flavonoid,which could react with ruthenium chloride to confirm that the ester bond of the compounds was at the 7-position hydroxyl group.Activity experiments showed that the activity of the compound with an amine-methylene group that containing a nitrogen atom at the 8-position is stronger than the lead compound.And after the ester bond is introduced at the 7-position,the activity of most compounds becomes better.

    Mechanism of Pinellia-Scutellaria-Coptis in treating Helicobacter pylori-related gastritis based on network pharmacology

    Consulting the previous research,the paper analyzed the main effective compounds and mechanisms of Pinellia, Scutellaria and Coptis in treating Helicobacter pylori associated gastritis based on network pharmacology.The compounds of Pinellia,Scutellaria and Coptis were obtained in the TCMSP platform.The targets of compounds were obtained in the Swiss Target Prediction database.The targets of Helicobacter pylori associated gastritis were obtained in the Gene Cards database and the OMIM database.Constructed a "herbs-compounds-targets" network association diagram and protein-protein interaction to analyze the main effective compounds and targets in Pinellia,Scutellaria and Coptis.A functional enrichment analysis and metabolic pathway of the targets of Pinellia,Scutellaria and Coptis were analyzed in David database.A total of 35 potential active compounds were screened,37 targets were related to disease and 77 pathways were involved.The herapeutic mechanism of Pinellia,Scutellaria and Coptis on the Helicobacter pylori  associated gastritis and regulating the gastric cancer reflects the characteristics of multi-component,multi-target,and multi-pathway,which provides a scientific basis for further explaining the action mechanism and the material basis of Pinellia,Scutellaria and Coptis against Helicobacter pylori associated gastritis.

    Screening of core genes and key microRNAs in the treatment of Helicobacter pylori related peptic ulcer by Scutellariae Radix and Coptidis Rhizoma

    The aim of this study was to explore the molecular network regulation mechanism of Coptidis Rhizoma and Scutellariae Radix (SRCR) on treatment of Helicobacter pylori (Hp) associated peptic ulcer (PU) based on network pharmacology and bioinformatics methods,and to screen out the core genes and key microRNAs.Firstly,the main active ingredients and possible targets of SRCR were retrieved through the Chinese Medicine System Pharmacology Database and Analysis Platform (TCMSP).GSE70394 gene expression profile data was downloaded from the GEO database.The microarray data of differentially expressed genes in Hp infected human gastric epithelial cells were homogenized by R software,and the volcano map and cluster heat map of differentially expressed genes were drawn.Secondly,STRING database was used to construct the protein interaction network between SRCR component target and Hp- infected human GES cells.The functions of GO and the enrichment of KEGG pathways of the above-mentioned characteristic genes were analyzed by DAVID database.Then,the core genes involved in the treatment of Hp related PU by SRCR were screened out by cytohubba plug-in unit.The key microRNAs regulating the core genes were predicted by using the TargetScan and miRBase databases.Finally,41 active components and 216 potential targets of SRCR were retrieved.The quality of GEO chip data was good.With P< 0.05 and︱logFC︱≥ 2 as the screening conditions,128 differentially expressed genes were obtained,including 77 up-regulated genes and 51 down-regulated genes.127 characteristic gene targets were further obtained for the treatment of HpPU by SRCR.Go analysis of these targets showed that the biological process involved 55 items such as inflammatory response,immune response,etc.Cell components included 11 items such as extracellular space,exosomes,nuclei,etc.Molecular functions included 15 items such as growth factor activity,epidermal growth factor receptor binding,etc.KEGG results showed that 33 signaling pathways,including cytokine receptor interaction pathway,nod like receptor signaling pathway,NF-κB signaling pathway,etc.were closely related to the treatment of Hp related PU by SRCR.Ten core genes regulated by SRCR,such as CXCL8,IL10,IL1B,etc.were screened out by cytohubba.Ten of the most functional microRNAs,such as mir-93-5p,mir-374b-5p,mir-3937,etc.which may be regulated by SRCR,were further screened out.The above results indicate that the treatment of HpPU with SRCR has the characteristics of multi-component,multi-target,and multi-channel synergy.SRCR can exert its therapeutic effect on HpPU by regulating the coding RNA (gene) and non-coding RNA (microRNA).

    Study on pathway and fermentation conditions of an endophytic fungus transforming baicalin to oroxylin A

    Aimed to provid a new approach for the exploitation of natural product oroxylin A,this reasearch focused on obtaining oroxylin A from baicalin by biotransformation applying endophytic fungus-Penicillium sp.R3 from Scutellaria baicalensis Georgi,identifying its biotransformation pathway and optimizing the fermentation conditions.The culture and biotransformation reaction were carried out in potato dextrose broth(PDB).Cell growth was measured by dry weight and the maximum biomass of Penicillium sp.R3 was gained on the day 4,then it entered the stationary phase.The biotransformation pathway was analyzed by the method of adding different substrates and the products were measured and identified by HPLC.After fermentation baicalin was converted to oroxylin A accompanied with baicalein which was identified as intermediate metabolite.Single factor analysis and orthogonal test were applied to optimized fermentation conditions.The optimal fermentation conditions were as follows:the liquid medium volume was 10% of the bottle;the concentration of inoculated spores was 1×106-2×106/mL and the temperature and shaker speed were 30 ℃,150 rpm respectively.The substrate baicalin was added on the day 3 with proportion of 0.08%,then the biotransformation process sustained 6 days.Under these optimal conditions the molar conversion rate and concentration of oroxylin A reached 42.44% and 177 mg/L respectively.The growth regularity and the biotransformation pathway of Penicillium sp.R3 was clarified,the stable and optimal conditions were obtained with a satisfactory yeild.All these findings lay a foundation for the further development and utilization of oroxylin A.

    The purpose of the study was to investigate the effect of different proportions of Scutellariae Radix(SR) and Fritillariae Cirrhosae Bulbus (FCB) on the content of main chemical components in SR and the anti-pneumonia effect of equal proportions.SR and FCB was mixed in the ratios of 3∶0,3∶1,3∶2,3∶3,2∶3,1∶3,0∶3,and then extracted with water and determined by HPLC to determine baicalin,wogonoside,baicalein and wogonin.Thirty-six healthy ICR mice were randomly divided into six groups:control group,LPS group,treatment group,SR group,FCB group,and SRFCB group,six in each group.The drug was administered for 6 consecutive days.On the 7th day,the experimental group was injected intraperitoneally with 10 mg/kg of LPS,and the control group was injected with an equal volume of normal saline.LPS was used for 6 hours to determine the levels of IL-1β,IL-6 and TNF-α in the serum of the mice.The pathological changes of lung tissue of mice in each group were observed under light microscope.The results showed that the total content of baicalin,wogonoside,baicallein,wogonin was less than 6% when the compatibility ratio were 3∶1,3∶2,3∶3 and 2∶3.When the compatibility ratio reached 1∶3,the total amount of the four chemical components decreased by 23.7%.After equal proportions compatibility of SR and FCB,the levels of IL-1β,IL-6,TNF-α in the serum of mice were significantly reduced,and the pathological morphology of lung tissue was improved,and the therapeutic effect was better than that of the single drug group.It is advisable to control the proportions compatibility of SR and FCB to 3∶1-2∶3,and they have a synergistic effect in anti-pneumonia.

    In this study,the inhibitory effect and mechanism of baicalein on tumor growth and lung metastasis of human breast cancer cell line MDA-MB-231 in nude mice was investigated.Twenty female BALB/C nude mice were randomly divided into normal group,model group,paclitaxel group and baicalein group.There were 5 nude mice in each group.The nude mice model of human breast cancer xenograft was established.Baicalein group was injected with 50 mg/kg baicalein abdominal cavity,paclitaxel group was injected with 10 mg/kg paclitaxel,normal group and model group were injected with normal saline for 8 weeks.Experimental results indicated that the tumor weight and the number of lung metastasis nodule and metastasis focus in paclitaxel group and baicalein group were less than those in the model group.Baicalein alone or in combination with ROCK inhibitor Y-27632 could inhibit the migration and cytoskeleton protein expression of MDA-MB-231 cells,reduce the formation of protein bundles,and inhibit the protein levels of ROCK1,Rac1,RhoA and p-MLC.

    In order to investigate the effect of wogonin on atherosclerosis and arterial inflammation in ApoE-/- mice fed with high fat diet,ApoE-/- mice were divided into normal diet group (ND),high-fat diet group (HFD),high-fat diet + wogonin (10 mg/kg) group (HFD + wogonin 10 mg/kg) and high-fat diet + wogonin (20 mg/kg) group (HFD + wogonin 20 mg/kg).Mice were given high-fat diet and wogonin for 12 weeks and then the arterial tissues were collected.RT-qPCR was used to detect the mRNA expression of IL-6,TNF-α,ICAM-1 and VCAM-1.Immunofluorescence staining was used to detect the expression of Moma-2 and VCAM-1 in the aortic root.Oil red O staining and HE staining were used to detect the changes of the lipid deposition and pathological changes in the aortic root.Biochemical kits were used to detect the serum lipid levels.Western blot was used to detect the phosphorylation of NF-κB and the expression of IκB-α.The results showed that compared with HFD group,the area of atherosclerotic plaque in HFD+wogonin group was decreased (P<0.05),but the serum lipid levels do not change; moreover,monocyte/monocyte macrophage infiltration,the protein expression of VCAM-1,the mRNA expression level of TNF-α,IL-6,ICAM-1 and VCAM-1 and the activation of NF-κB signal pathway in HFD+wogonin group were significantly reduced (P<0.05).These findings indicated that wogonin could effectively alleviate arterial atherosclerosis and inhibit NF-κB-mediated arterial inflammation in ApoE-/- mice induced by high-fat diet.
