亳产桔梗提取物对非小细胞肺癌细胞增殖及转移的影响及机制研究

Effect and mechanism of Bozhou Platycodonis Radix extract on proliferation and metastasis of non-small cell lung cancer cells

摘要:

探讨亳产桔梗提取物(Platycodonis Radix extract,PRE)对非小细胞肺癌(non-small cell lung cancer,NSCLC)细胞的作用及机制。实验中,用不同浓度PRE处理人肺癌细胞A549和人肺成纤维细胞HLF,通过MTT试验、克隆形成实验、细胞划痕实验和Transwell实验,评估其对细胞增殖、迁移和侵袭能力的影响,同时采用Western blot检测上皮间质转化(epithelial-mesenchymal transition,EMT)及Ras同源物基因家族成员A(Ras homolog gene family, member A,RhoA)/Rho相关卷曲螺旋形成蛋白激酶(Rho-associated coiled-coil forming kinase,ROCK)通路相关蛋白的表达。此外,还通过裸鼠移植瘤实验,观察不同剂量PRE及溶血磷脂酸(lysophosphatidic acid,LPA)注射对肿瘤体积和重量的影响。结果显示,PRE能显著抑制NSCLC细胞的活力、克隆数、迁移率和穿模细胞数,下调相关蛋白表达,且呈剂量依赖性;而经LPA干预后则呈现相反效果。裸鼠实验也进一步证实了PRE的抑制作用。因此,PRE可能通过抑制RhoA/ROCK信号通路和EMT过程,发挥抗NSCLC作用。

Abstract:

This study aimed to investigate the effects and mechanisms of Bozhou Platycodonis Radix extract (PRE) on non-small cell lung cancer (NSCLC) cells. In the experiments, human lung cancer cell line A549 and human lung fibroblast cell line HLF were treated with different concentrations of PRE.. The impacts on cell proliferation, migration, and invasion abilities were evaluated through MTT assay, colony formation assay, cell wound-healing assay, and Transwell assay. Western blot was used to detect the expression of epithelial-mesenchymal transition (EMT) and Ras homolog gene family, member A (RhoA)/Rho-associated coiled-coil forming kinase (ROCK) pathway-related proteins. Additionally, a nude mouse xenograft tumor experiment was conducted to observe the effects of different doses of PRE and lysophosphatidic acid (LPA) injection on tumor volume and weight. The results showed that PRE could significantly inhibit the viability, colony number, migration rate, and the number of cells passing through the membrane of NSCLC cells, and down-regulate the expressions of related proteins in a dose-dependent manner. In contrast, LPA intervention produced the opposite effects. The nude mouse experiment further confirmed the inhibitory effect of PRE. Therefore, PRE may exert anti-NSCLC effects by inhibiting the RhoA/ROCK signaling pathway and the EMT process.
