姜黄素对高脂喂养大鼠肝脏脂肪变性的影响及机制研究

摘要:

探讨姜黄素对高脂喂养大鼠肝脏脂肪变性的影响及机制。将SD大鼠随机分为正常对照(normal control,NC)组、高脂喂养(high-fat diet,HF)组、姜黄素低剂量(low-dose curcumin,Cur-L)组和姜黄素高剂量(high-dose curcumin,Cur-H)组。测定大鼠Lee's指数和肝指数;HE染色和油红O染色观察肝脏脂肪变性的情况;观察大鼠血甘油三酯(triglyceride,TG)和丙氨酸氨基转移酶(alanine aminotransferase,ALT)以及肝脏TG和游离脂肪酸(free fat acid,FFA)水平;Western blot法检测肝组织哺乳动物雷帕霉素靶蛋白(mammalian target of rapamycin,mTOR)、固醇调节元件结合蛋白1c(sterol regulatory element binding protein-1c,SREBP-1c)、脂肪酸合成酶(fatty acid synthase,FAS)蛋白的表达。HF组大鼠Lee's指数和肝指数较NC组明显升高(P<0.05);Cur-L及Cur-H组大鼠Lee's指数和肝指数较HF组明显降低(P<0.05)。NC组大鼠肝脏无脂肪变,HF组大鼠肝脏脂肪变性明显,应用姜黄素后肝脏脂肪变性明显减轻,且有剂量依赖性。与NC组相比,HF组大鼠血TG和ALT以及肝脏TG和FFA水平均明显升高(P<0.05),肝组织mTOR、SREBP-1c和FAS蛋白表达明显升高(P<0.05)。姜黄素干预后,大鼠血TG和ALT以及肝脏TG和FFA水平均明显下降,且有剂量依赖性(P<0.05);与HF组相比,姜黄素组大鼠肝组织mTOR、SREBP-1c和FAS蛋白表达明显降低,且有剂量依赖性(P<0.05)。研究结果表明,姜黄素可能通过降低mTOR和SREBP-1c的表达减轻高脂喂养大鼠肝脏脂肪变性。

Abstract:

This study aims to explore the effects and mechanisms of curcumin on hepatic steatosis in high-fat-fed rats. SD rats were randomly divided into three group: normal control (NC) group, high-fat diet (HF) group, low-dose curcumin (Cur-L) group, and high-dose curcumin(Cur-H) group. The Lee's index and liver index were measured. Fatty changes in the liver observed by HE and oil red O staining. The levels of triglyceride (TG) and alanine aminotransferase (ALT) in blood and the levels of TG and free fat acid (FFA) in liver were detected. The expression of mammalian target of rapamycin (mTOR), sterol regulatory element binding protein-1c (SREBP-1c) and fatty acid synthase (FAS) protein in liver was detected by Western blot. The Lee's index and liver index in HF group were obviously increased than those in NC group (P<0.05). The Lee's index and liver index in Cur-L and Cur-H group were significantly lower than those in HF group (P<0.05). There was no hepatic steatosis in NC group. In HF group, hepatic steatosis was obvious. Hepatic steatosis was significantly reduced after the application of curcumin, and there was a dose-dependent difference. Compared with NC group, the levels of TG and ALT in blood and TG and FFA content of liver in HF group were obviously increased (P<0.05); The expression of mTOR, SREBP-1c and FAS protein in liver was markedly elevated (P<0.05). After the application of curcumin, the levels of TG and ALT in blood and TG and FFA content of liver were decreased significantly, and there was a dose-dependent difference (P<0.05); Compared with HF group, the expression of mTOR, SREBP-1c and FAS protein in liver was significantly reduced, and there was a dose-dependent difference (P<0.05). Curcumin may alleviate liver steatosis in high-fat fed rats by reducing the expression of mTOR and SREBP-1c.
