基于网络药理学研究中药复方及其联合水解酪蛋白肽对自发性高血压大鼠的降压作用及机制

摘要:

该文基于网络药理学预测及实验验证的方法,对中药复方联合水解酪蛋白肽降高血压的作用机制进行了探讨。在TCM-ID数据库、TCMID数据库、TCM@TAIWAN数据库中筛选中药组方及水解酪蛋白肽的主要活性成分,选取芦丁、槲皮素、表儿茶素、原花青素B2、αs1-CN、αs2-CN作为主要活性成分用于网络药理学分析,筛选主要成分作用于心血管疾病的网络靶点,结合KEGG和Uniprot数据库,进行信号通路和生物途径分析,预测其作用机制。在网络药理学预测的基础上,建立体外动物模型,观察及研究中药复方联合水解酪蛋白肽对自发性高血压大鼠的降压作用及机制。网络分析结果显示,组方可通过作用同一靶点或不同靶点发挥抗高血压作用,其作用机制与调节肾素-血管紧张素系统、血管内皮紧张因子-收缩因子系统等通路,参与炎症反应、内皮细胞保护及脂质生物合成和代谢等生物途径有关。即在动物模型上,组方主要通过cGMP-PKG信号通路,升高NO、AngII水平,降低ET-1、ALD、TNF-α水平达到降低血压、增强免疫、保肾等作用,验证了网络药理学的预测结果,综合阐释了组方抗高血压的作用机制。

Abstract:

Based on the method of network pharmacology prediction and experimental verification,this article discusses the mechanism of reducing hypertension of Chinese herbal compound combined with hydrolyzed casein peptide.The main active ingredients of traditional Chinese medicine prescriptions and hydrolyzed casein peptides in TCM-ID database were screened.TCMID database,TCM@TAIWAN database,and select rutin,quercetin,epicatechin,procyanidin B2,αs1-CN,αs2-CN were used as the main active ingredient for network pharmacological analysis to screen the network targets of the main ingredients acting on cardiovascular diseases;combined with KEGG and Uniprot database,signal pathway and biological pathway analysis weer conducted to predict its mechanism of action.Based on network pharmacology prediction,an in vitro animal model was established to observe and study the antihypertensive effect and mechanism of Chinese herbal compound combined with hydrolyzed casein peptides on spontaneously hypertensive rats.The results of the network analysis showed that the prescriptions can exert anti-hypertensive effects by acting on the same target or different targets,and its mechanism of action was related to the regulation of the renin-angiotensin system,vascular endothelial tension factor-contraction factor system and other pathways,participating in the inflammatory response,endothelial cell protection and lipid biosynthesis and metabolism and other biological pathways.In other words,in animal models,by acting cGMP-PKG signaling athway,the prescription increased NO and AngII levels,and reduced ET-1,ALD,and TNF-α levels to reduce blood pressure,enhance immunity,and protect kidneys and so on,which verified the prediction results of network pharmacology,and comprehensively explained the antihypertensive mechanism of the prescription.
