硬尖神香草化学成分及其抗炎活性研究

Chemical constituents of Hyssopus cuspidatus Boriss. and their anti-inflammatory activity

摘要:

研究维药硬尖神香草的化学成分。采用硅胶、凝胶、ODS等柱色谱结合半制备液相色谱技术进行分离纯化,并通过现代波谱学技术对化合物进行结构鉴定。同时采用MTT法检测了所得化合物的抗炎活性。从维药硬尖神香草95%乙醇提取物中共分离得到18个化合物,分别鉴定为:(1S,2R,5S)-2-羟基-2-(羟甲基)-6,6-二甲基双环[3,1,1]庚-3-酮(1)、19,20-环氧-12-甲氧基-11,14,19-三羟基-7-氧代-8,11,13-松香三烯(2)、(1R,1′R,5S,5′S)-6,6,6′,6′-四甲基[2,2′-二环[3,1,1]庚烷]-2,2′-二醇(3)、4-羟基-6,6-二甲基双环[3,1,1]庚-2-烯-2-甲酸甲酯(4)、4-表阿诺醇(5)、β-谷甾醇(6)、熊果酸(7)、2,3-二氢-6,7-二甲氧基-2-甲基-1H-茚-1-酮(8)、(2R)-5-羟基-7,2',3'-三甲氧基黄酮(9)、4'-羟基-5,6,7-三甲氧基黄酮(10)、佛手柑内酯(11)、羟基二氢博伏内酯(12)、蒎酸(13)、(E)-3-(4-羟基苯基)丙烯酸乙酯(14)、阿魏酸(15)、1-阿魏酸甘油酯(16)、丁香醇(17)、水杨酸(18)。化合物3~589121417为首次从硬尖神香草中分离得到。化合物2~49具有较为显著的抗炎活性,其对脂多糖诱导的RAW 264.7细胞释放NO的IC50值为0.15~0.39 μg/mL。

Abstract:

This study aims to investigate the chemical constituents of the Uyghur medicinal herb Hyssopus cuspidatus Boriss. Isolation and purification were carried out using column chromatography techniques such as silica gel, Sephadex LH-20, and ODS, combined with semi-preparative HPLC. The structures of the isolated compounds were elucidated based on extensive spectroscopic analysis. In addition, the anti-inflammatory activities of the obtained compounds were evaluated using the MTT assay.  Eighteen compounds were isolated from the 95% ethanol extract of H. cuspidatus and identified as: (1S, 2R, 5S)-2-hydroxy-2-(hydroxymethyl)-6, 6-dimethylbicyclo [3, 1, 1] heptan-3-one (1), 19, 20-epoxy-12-methoxy-11, 14, 19-trihydroxy-7-oxo-8, 11, 13-abietatriene (2), (1R, 1′R, 5S, 5′S)-6, 6, 6′, 6′-tetramethyl [2, 2′-bicyclo [3, 1, 1] heptane]-2, 2′-diol (3), methyl 4-hydroxy-6, 6-dimethylbicyclo [3, 1, 1] hept-2-ene-2-carboxylate (4), 4-epianol (5), β-sitosterol (6), ursolic acid (7), 2, 3-dihydro-6, 7-dimethoxy-2-methyl-1H-inden-1-one (8), (2R)-5-hydroxy-7, 2', 3'-trimethoxyflavone (9), 4'-hydroxy-5, 6, 7-trimethoxyflavone (10), bergapten (11), hydroxydihydrobovolide (12), pinic acid (13), Ethyl (E)-3-(4-hydroxyphenyl)acrylate (14), ferulic acid (15), 1-feruloylglycerol (16), syringyl alcohol (17), salicylic acid (18). Compounds 3-5, 8, 9, 12, 14 and 17 are isolated from H. cuspidatus for the first time. Compounds 2-4 and 9 exhibited significant anti-inflammatory activity, with IC50 values of 0.15-0.39 μg/mL against nitric oxide release in lipopolysaccharide-induced RAW 264.7 cells.
