茶芎苯酞类成分抗缺血性脑卒中机制及药动学研究进展

Research progress on mechanism and pharmacokinetics of Ligusticum sinense against cerebral ischemic stroke

摘要:

茶芎是江西省九江地区的特产中药材之一,又名抚芎,系伞形科藁本属植物茶芎的干燥根茎,有行气活血、祛风止痛功效。作为一种民间药食两用中药,茶芎中苯酞类成分具有多种药理作用,其良好的抗缺血性脑卒中(cerebral ischemic stroke,CIS)作用近年来受到越来越多关注。现代研究表明,茶芎中苯酞类化合物能通过抑制神经细胞炎症反应、调节神经元兴奋毒性、保护血脑屏障,减轻神经细胞氧化应激、抑制神经细胞凋亡与自噬、舒张和扩张血管等,缓解治疗CIS,多成分、多靶点、多途径治疗CIS。了解茶芎抗CIS的药效成分在体内外的作用机制及药动学特征可更好指导其临床用药安全规范和制剂开发。本文通过查阅国内外文献,系统总结了茶芎苯酞类成分组成及药效成分抗CIS作用机制及药动学研究并进行归纳综述,以期为茶芎抗CIS的深入研究和新药研发提供参考依据。

Abstract:

Ligusticum sinense, also named Fuxiong, is one of the specialty Chinese medicinal herbs in Jiujiang, Jiangxi Province. It is the dried rhizome of L. sinense, a plant belonging to the Umbelliferae family and the Ligusticum genus. It has the effects of moving qi and activating blood, expelling wind and relieving pain. As a traditional Chinese medicine that can be used as both medicine and food, the phthalein in L. sinense has various pharmacological effects, and it’s excellent anti-cerebral ischemic stroke (CIS) effect has received increasing attention in recent years. Modern research has shown that phthalides compounds in L. sinense can alleviate the treatment of CIS by inhibiting neuronal inflammation response, regulation neuronal excitotoxicity, protecting the blood-brain barrier, reducing neuronal oxidative stress, inhibiting neruonal apoptosis and autophagy, and dilating blood vessels, etc. It is a multipathway treatment for CIS. Understanding the mechanism of action and pharmacokinetic characteristics of the active ingredients of L. sinense against cerebral ischemic stroke in vitro and in vivo can better guide its clinical medication safety standards and formulation development. This article systematically summarizes the composition and pharmacological mechanism of the phthalide components in L. sinense and their anti CIS effects, as well as pharmacokinetic studies, by reviewing domestic and foreign literature. The aim is to provide a reference for further research and new drug development of L. sinense in the treatment of CIS.
