天山棱子芹对胰脂肪酶、α-葡萄糖苷酶和血管紧张素转化酶的抑制作用研究

Inhibitory effect of Pleurospermum lindleyanum on pancreatic lipase, α-glucosidase and angiotensin-converting enzyme

摘要:

探究天山棱子芹及其活性成分对胰脂肪酶、α-葡萄糖苷酶及血管紧张素转化酶的抑制作用。本文通过大孔吸附树脂纯化天山棱子芹提取物,并测定天山棱子芹及其活性成分对3个酶的活性抑制率。结果表明,天山棱子芹醇提物中绿原酸、木犀草苷、木犀草素、佛手苷内酯、氧化前胡素5个活性成分含量均高于水提物。且经纯化后,纯化物中各活性成分含量均有明显提高。天山棱子芹纯化物对3个酶的抑制作用最好,IC50值为0.594、0.469、0.746 mg/mL。活性成分中,绿原酸和木犀草素对3个酶均有明显的抑制作用,IC50值为0.145、0.131、0.136、0.159、0.134、0.140 mg/mL,氧化前胡素对胰脂肪酶和血管紧张素转化酶有较好的抑制作用,IC50值为0.392、0.152 mg/mL,木犀草苷对胰脂肪酶、佛手苷内酯对血管紧张素转化酶有一定的抑制作用,IC50值为0.225、0.258 mg/mL。因此天山棱子芹具有良好的降脂降糖降压活性,可为天山棱子芹防治代谢综合征提供理论依据。

Abstract:

A study was conducted to investigate the inhibitory effects of Pleurospermum lindleyanum and its active components on pancreatic lipase, α-glucosidase, and angiotensin-converting enzyme. In this paper, the extract of P. lindleyanum was purified by macroporous adsorption resin, and the activity inhibition rates of P. lindleyanum and its active components against the three enzymes were determined. The results showed that the contents of five active components, chlorogenic acid, luteolin-7-O-glucoside, luteolin, bergapten, and oxypeucedanin, in the ethanol extract of P. lindleyanum were all higher than those in the aqueous extract. After purification, the contents of each active component in the purified product were significantly increased. The best inhibitory effects on the three enzymes were exerted by the purified product of P. lindleyanum, with IC50 values of 0.594, 0.469 and 0.746 mg/mL. Among the active components, significant inhibitory effects on all three enzymes were exhibited by chlorogenic acid and luteolin, with IC50 values of 0.145, 0.131, 0.136, 0.159, 0.134 and 0.140 mg/mL. Good inhibitory effects on pancreatic lipase and angiotensin-converting enzyme were shown by oxypeucedanin, with IC50 values of 0.392 and 0.152 mg/mL. Respectively, moderate inhibitory effects on pancreatic lipase and angiotensin-converting enzyme were exhibited by luteolin-7-O-glucoside and bergapten, with IC50 values of 0.225 and 0.258 mg/mL. Therefore, P. lindleyanum was demonstrated to possess good lipid-lowering, hypoglycemic and antihypertensive activities, which could provide a theoretical basis for the prevention and treatment of metabolic syndrome by P. lindleyanum.
