姜黄素对大鼠运动性心肌纤维化的保护作用

Protective effect of curcumin on exercise-induced myocardial fibrosis of rats

摘要:

本文就姜黄素调控p38 MAPK/NF-κB信号通路对长时程、大强度运动致大鼠运动性心肌纤维化的保护作用进行了研究。SPF级雄性SD大鼠随机分为对照组(control group,C)、模型组(model group,M)、姜黄素+模型组(curcumin+model group,CM)。C组安静饲养,其余组进行6周递增负荷跑台训练。训练期间CM组灌胃姜黄素,其余组灌胃等体积0.5%羧甲基纤维素钠。训练后24 h取材,观察心肌组织形态与超微结构,检测血清、心肌相关生化指标。结果显示,心肌组织形态,M组较C组出现损伤性改变,心肌纤维出现断裂,且伴有炎症浸润、结缔组织增生现象,CM组较M组改善明显;心肌超微结构,M组线粒体嵴短缩,部分出现空泡变性,CM组无明显异常。与C组相比,M组心肌组织糖原容积分数、血清cTNI及心肌TGF-β1、TNF-α、IL-1β、p-p38 MAPK和NF-κB p65显著上升(P<0.01);与M组相比,CM组心肌组织糖原容积分数、血清cTNI及心肌TGF-β1、TNF-α、IL-1β、p-p38 MAPK和NF-κB p65显著下降(P<0.05或P<0.01)。从而说明,姜黄素可能通过抑制p38 MAPK/NF-κB信号通路相关蛋白质过度活化和表达,调控心肌组织炎症因子合成与分泌及细胞外基质(extracellular matrix,ECM)合成与降解间的平衡,缓解心肌纤维化。

Abstract:

The protective effect of curcumin on p38 MAPK/NF-κB signaling pathway on myocardial fibrosis induced by prolonged and intensity exercise in rats was studied.SPF male SD rats were randomly divided into control group (C),model group (M),and curcumin + model group (CM).Group C received quiet feeding,while the other groups received increasing load treadmill training for 6 weeks.During the period,group CM was given a gavage of curcumin,while the other groups were given a gavage of with 0.5% sodium carboxymethyl cellulose of the same volume.The samples were taken to observe the morphology and ultrastructure of the myocardium,and detect the serum and myocardial biochemical indexes 24 h after the training.The results showed that compared with group C in myocardial tissue morphology,there were injurious changes,myocardial fiber fracture,inflammatory infiltration and connective tissue hyperplasia in group M,the improvement was more obvious in group CM than that in group M;myocardial ultrastructure,mitochondrial cristae of group M shortened,part of them appeared vacuolar degeneration,there was no obvious abnormality in group CM.Compared with group C,glycogen volume fraction of myocardial tissue,cTNI,TGF-β1,TNF-α,IL-1β,p-p38 MAPK and NF-κB p65 in myocardium increased significantly(P<0.01);Compared with group M,glycogen volume fraction of myocardial tissue,cTNI,TGF-β1,TNF-α,IL-1β,p-p38 MAPK and NF-κB p65 in myocardium decreased significantly(P<0.05 or P<0.01).These results suggested that curcumin may inhibit the over-activation and expression of the related proteins in p38 MAPK/NF-κB signaling pathway,balance the synthesis and secretion of inflammatory factors,and the synthesis and degradation of ECM in myocardial tissue,then alleviate myocardial fibrosis.
