Determination of Anthraquinone Derivatives in Rhubarb Seeds

Abstract: The aim of this study was to explore the concentrations of anthraquinone derivatives detected in Rheum tanguticum Maxim.ex Balf.seeds, R.palmatum Linn.seeds and R.officinale Baill.seeds.The contents of aloe-emodin,rhein,emodin,emodin,chrysophanol in rhubarb seeds were determined by highperformance liquid chromatography (HPLC).Agilent Eclipse plus C18 (4.6×250 mm,5 μm) column was used for chromatographic separation with the mobile phase consisting of methanol (A) and 0.1% acetic acid (B) in a gradient elution mode.Column temperature was 25 ℃;Detection wavelength was 254 nm.The results showed that the order of content of concentrations of anthraquinone derivatives was Rh palmatum Linn.seeds > Rheum tanguticum Maxim.ex Balf.seeds > Rh.officinale Baill.seeds.Rhubarb seeds contain abundant emodin,emodin and chrysophanol,excluding aloe-emodin and rhein.The developed method was proved to be accurate,precise,stable and suitable for the determination of anthraquinone derivatives in Rhubarb seeds.
