Simultaneous Determination of 6 Active Compounds in Different Habitats and Different Parts of Tripterygium hypoglaucum Hutch by UPLC-MS

Abstract: An ultra-performance liquid chromatography-mass spectrometry (UPLC-MS) method was developed for the simultaneous determination of wilforgine,wilforine,triptolide,demethylzeylasteral,triptophenolide and wilforlide A in different habitats and different parts of Tripterygium hypoglaucum Hutch.The chromatographic separation was performed on a Phenomenex Luna C18 column (50×2.0 mm,3 μm) with gradient elution consisting of acetonitrile and 0.1% formic acid in water at a flow rate of 0.3 mL/min.The column temperature was set at 30 ℃.Detection was carried out on a triple quadrupole tandem mass spectrometry by multiple-reaction monitoring (MRM) and an electrospray ionization (ESI) was operated in positive mode.All calibration curves of the 6 active compounds showed good linearity in the range of 6.80-1360.00,5.90-1180.00,1.20-240.00,25.05-5010.00,1.45-290.00 and 1.65-330.00μg/L (r≥0.9998).The recoveries were in the range of 98.19%-100.91% and RSDs were between 1.46% and 2.99%.The method was applied for the determination of 6 compounds in 36 samples of T.hypoglaucum Hutch from different habitats and different parts.The results indicated that this method was simple,rapid and reliable.It can be used for the quality control of T.hypoglaucum Hutch.
