Regulation Effect of Allicin on Beta Secretase Subunit Bace1 Mediated by ERK1/2 Signaling Pathway

Abstract: In this study,the regulation effect of ERK1/2 signal pathway mediated allicin on beta secretase subunit Bace1 was investigated by immunofluorescence,Western blot,Real-Time PCR and ELISA methods.It was found that the Bace1 expression in SH-SY5Y cells which treated with allicin was lower than control group,but treated with allicin and ERK1/2 pathway inhibitors PD98059,the expression of Bace1 was rose.The results showed that allicin can down-regulate Bace1 expression through ERK1/2 pathway.ELISA results showed that the expression of A beta 1-40 and A beta 1-42 were decreased in SH-SY5Y cells which treated with allicin,while the expressions were increased in SH-SY5Y cells which treated with allicin and PD98059,compared with allicin treatment group.These results prompted that allicin can inhibit the beta secretase activity,downregulate Bace1 expression,and suggested that allicin had protective effect on neurons by decreasing the production of Aβ.
