Inhibitory effect of evodiamine combined with gemcitabine on human tongue squamous cell carcinoma in nude mice and its mechanism

Abstract: The objective of this study was to investigate the inhibitory effect of evodiamine combined with gemcitabine on subcutaneous xenograft of tongue squamous cell carcinoma in nude mice and its mechanism.The nude mice model of tongue squamous cell carcinoma was established.The tumor volume,food intake,water intake and response to external stimuli were observed after drug intervention.HE staining was used to observe the pathological structure of the transplanted tumor and various organs.Transmission electron microscope was used to observe the tumor tissue ultrastructure,western blot and immunohistochemistry were used to detect the expression of NF-κB and Bcl-2 family proteins.The results showed that compared with the model control group and the single drug group,the volume of transplanted tumors in the combined group was significantly reduced,the quality of life of the tumor-bearing nude mice was improved,the number of pathological mitotic figures in the tumor cells was decreased,and the apoptotic rate was increased.The expression levels of Bax and Ik-βα were significantly increased,and the expression of Bcl-2,Bcl-xl and P65 were significantly decreased.The antitumor mechanism of evodiamine combined with gemcitabine may be related to the regulation of NF-κB and Bcl-2 family proteins.
