Study on pharmacokinetics and metabolites of a kaempferol glycoside from Hippophae rhamnoides subsp.sinensis in vivo

Abstract:

Kaempferol-3-3-O-β-D-sophoroside-7-O-α-L-rhamnoside (KSR) is isolated from Hippophae rhamnoides subsp. sinensis.In this study,in order to better study the active constituents in vivo,the ADME (absorption,distribution,metabolism and excretion) of KSR were also studied by LC-MS/MS.In the pharmacokinetic analysis,KSR exhibited high elimination (T=4-6 h) and poor absolute bioavailability (0.33%± 0.09%) after oral administration.In the tissue distribution,the content of KSR was highest in liver tissue.In excretion,KSR was excreted in the form of detracting 7-O-rhamnose from the body.In this study,we initially elucidated the pharmacokinetics and metabolites of KSR,which may have an important significance to study flavonoids in Hippophae rhamnoides L.
