Simultaneous Determination of Betulinic Acid,Oleanolic Acid and Ursolic Acid in Persimmon Leaves by RP-HPLC

Abstract: A RP-HPLC method was established for the simultaneous determination of betulinic acid,oleanolic acid and ursolic acid in persimmon leaves.Thermo C18 column (250 mm × 4.6 mm,5 μm)was used for the chromatographic separation,acetonitrile-phosphoric acid aqueous solution(pH=3.0)(85:15)was used as the mobile phase with a flow rate of 0.8 mL/min.The detection wave length was 209 nm and the temperature of the column was 30 ℃.The linear ranges for betulinic acid,oleanolic acid and ursolic acid were 0.124-3.100 μg(r=0.9996),0.160-4.000 μg(r=0.9994)and 0.288-7.200 μg(r=0.9998),respectively.The average recoveries of the developed HPLC method were 98.22% for betulinic acid,95.23% for oleanolic acid and 103.87% for ursolic acid.The content of betulinic acid,oleanolic acid and ursolic acid in the leave of MoPan were 0.872,1.406 and 4.598 mg/g,respectively.The developed method can be used to simultaneously determine the content of betulinic acid,oleanolic acid and ursolic acid in persimmon leaves.
