Determination of Tripterine in Tripterygium hypoglaucum by Ionic Liquidbased Ultrasoundassisted Extraction Combined with RPHPLC

Abstract: Using six kinds of ionic liquids as extractants,a method of ultrasonicassisted extraction combined with HPLC was developed for the simultaneous determination of tripterine in Tripterygium hypoglaucum.The separation was performed on an Inertsil ODS4 column with the mobile phase of acetonitrile0.1% phosphoric acid (80:20),detection wavelength was 425 nm and column temperature was 30 ℃.Under the optimal extractions,the results showed that tripterine had the highest extraction yield through using 0.6 mol·L1 [BMIm]PF6 methanol solution as extraction solvent with the solidliquid ratio of 1:4 (g·mL1).The calibration curves showed a good linearity in the range of 0.015045 μg (r=0.999 9),with the average recovery of 99.25% and RSD is 2.22%.These results indicated that this method is convenient,accurate,reliable and environment friendly,providing a reliable analytical method for the quality control of Tripterygium hypoglaucum.
