Purification Technology of Malus micromalus Makino Flavonoids by Macroporous Absorbent Resins

Abstract: The suitable resin was selected by static and dynamic adsorption from 10 kinds of macroporous absorbent resins to Malus micromalus Makino flavonoids. The purification technology of the crude extracts by macroporous adsorbing resins were decided through the experiments. The results showed that NKA-9 resin was selected because it gave the most effective of separation. The optimum process for the purification was as follows: the concentration of sample solution was 5.15 mg/mL, pH was 4.0, the loading weight was 4BV and the adsorption flow rate was 2 BV/h; then eluted with 80% ethanol for 3 BV with elution flow rate of 1BV/h. In these optimum conditions, average adsorption rate, desorption rate, yield and purity of the flavonoids was reached to (79.39±0.13)%, (84.14±0.11)%, (68.20±0.15)% and (28.81±0.06)% (n=5), respectively.
