沙棘汁对高盐饮食大鼠血压和心功能的作用研究

摘要:

探究沙棘汁对高盐饮食大鼠血压和心功能的影响及潜在的作用机制。将SD大鼠随机分为对照组、高盐组和沙棘汁组,实验第0、1、3、5、7周利用尾套法监测尾动脉血压,8周后计算心脏指数,左心室导管术检测左心室血流动力学变化,天狼星红染色观察心肌纤维化,Western blot检测心脏炎症相关蛋白NOD样受体热蛋白结构域相关蛋白3(NOD-like receptor pyrin domain-containing protein 3,NLRP3)、白介素-1β(interleukin-1β,IL-1β)、环氧合酶-2(cyclooxygenase-2,COX-2)的表达;ELISA检测血清中心肌损伤标志物肌红蛋白(myoglobin,MYO)、超敏心肌肌钙蛋白(high-sensitivity cardiac troponin,hs-cTn),血管内皮损伤标志物内皮素-1(endothelin-1,ET-1)、一氧化氮(nitric oxide,NO),血管内皮炎症标志物可溶性细胞间粘附分子-1(soluble intercellular adhesion molecule-1,sICAM-1)和可溶性血管细胞黏附分子-1(soluble vascular cell adhesion molecule,sVCAM-1),以及炎症因子超敏C反应蛋白(high-sensitivity C-reactive protein,hs-CRP)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、IL-1β的变化。结果表明,沙棘汁组大鼠收缩压、舒张压和平均动脉压均降低,心脏指数下降,左心室舒张末期压力降低,而左心室内压最大上升速率和最大下降速率绝对值升高,心脏天狼星红染色面积减少,NLRP3、IL-1β、COX-2蛋白表达降低,血清MYO、hs-cTn、ET-1、sICAM-1、sVCAM-1、hs-CRP、TNF-α、IL-1β含量降低,NO含量升高。综上所述,沙棘汁通过抑制炎症反应减轻血管内皮和心肌损伤,从而降低血压、改善心功能。

Abstract:

This study aims to investigate the effects of seabuckthorn juice (SBJ) on blood pressure and cardiac function in rats fed with high-salt diet, as well as the underlying mechanism. SD rats were randomly allocated into three groups: control group, high-salt diet group, and SBJ group. At 0, 1, 3, 5, and 7 weeks of the experiment, tail artery blood pressure was monitored using the tail-cuff method. After eight weeks, the cardiac index was calculated, left ventricular catheterization was performed to measure hemodynamic changes in the left ventricle, and Sirius Red staining was utilized to observe myocardial fibrosis, expressions of the inflammation-related proteins NOD-like receptor pyrin domain-containing protein 3 (NLRP3), interleukin-1β (IL-1β) and cyclooxygenase-2 (COX-2) in the heart were examined using Western blot, ELISA was employed to detect myocardial injury markers including myoglobin (MYO), high-sensitivity cardiac troponin (hs-cTn), vascular endothelial injury markers like endothelin-1 (ET-1), nitric oxide (NO), vascular endothelial inflammation markers like soluble intercellular adhesion molecule-1 (sICAM-1), soluble vascular cell adhesion molecule (sVCAM-1), and inflammatory factors such as high-sensitivity C-reactive protein (hs-CRP), tumor necrosis factor-α (TNF-α), and IL-1β in serum. The findings revealed that in the SBJ group, systolic blood pressure, diastolic blood pressure, and mean arterial pressure decreased, along with reduction in the cardiac index and left ventricular end-diastolic pressure. Nevertheless, the maximum rate of increase in left ventricular pressure and the absolute value of the maximum rate of decrease in left ventricular pressure increased. Furthermore, the Sirius Red staining area of heart diminished, and the protein expression levels of NLRP3, IL-1β, COX-2 decreased. Concurrently, the serum contents of MYO, hs-cTn, ET-1, sICAM-1, sVCAM-1, hs-CRP, TNF-α, and IL-1β exhibited a decline, while the NO content demonstrated an increase. In conclusion, SBJ mitigates vascular endothelial and myocardial injury by suppressing inflammatory responses, consequently reducing blood pressure and enhancing cardiac function.
