红豆树枝条化学成分及抗炎活性研究

Chemical constituents and their anti-inflammatory activity from twigs of Ormosia hosiei Hemsl.& E.H.Wilson

摘要:

采用D101大孔吸附树脂、硅胶、凝胶和制备高效液相色谱等柱色谱方法,对红豆树枝条70%乙醇水提取物进行系统分离,并结合现代波谱学手段对分离的化学成分进行结构鉴定,从中分离鉴定11个黄酮及其苷类化合物。分别为3′,4′,7-三羟基-5-甲氧基异黄酮(1)、3′,4′,5,7-四羟基二氢异黄酮(2)、染料木苷-5,4′-二甲醚(3)、澳白檀苷(4)、鹰嘴豆芽素A-7-O-芸香糖苷(5)、染料木苷(6)、芹菜素-4′-O-β-D-吡喃葡萄糖苷(7)、牡荆素(8)、江户樱花苷(9)、柚皮苷(10)、新北美圣草苷(11)。其中化合物1为新化合物,化合物2为新的天然产物,除化合物6外,其余均为首次从红豆属植物中分离。抗炎活性评价结果显示:当浓度为20 μmol/L时,化合物3、49对脂多糖诱导的小鼠BV-2小胶质细胞分泌白介素-6的抑制率分别为37.67%±3.27%、36.02%±5.48%和50.10%±1.58%。

Abstract:

A new isoflavones with ten known flavonoids were isolated from 70% ethanol extract of twigs of Ormosia hosiei by using various chromatographic techniques.Their structures were identified as 3′,4′,7-trihydroxy-5-methoxy isoflavone(1),3′,4′,5,7-tetrahydroxy dihydroisoflavone(2),genistin-5,4′-methyl ether(3),lanceolarin(4),biochanin A-7-O-rutinoside(5),genistin(6),apigenin-4′-O-β-D-glucoside(7),vitexin(8),prunin(9),naringin(10),neoeriocitrin(11).Among them,compound 1 was a new compound,compound 2 was a new natural product.All compounds expect compound 6 were isolated from the plants of the genus Ormosia for the first time.In addition,the evaluation results of anti-inflammatory activity showed that the inhibited IL-6 secretion from LPS-induced BV-2 microglia by compounds 3, 4 and 9 were 37.67%±3.27%,36.02%±5.48% and 50.10%±1.58%,at the concentration of 20 μmol/L.
