Optimization on Extraction Technology and Determination of Mulberroside A in Morus alba (L.) by UPLC

Abstract: Response Surface Methodology (RSM)was applied to optimize of ultrasonic extracting technique for mulberroside A in Morus alba (L.).On the basis of a single factor test,the effects of the proportion of methanol concentration,the volume of solvent,the extraction time and the extraction temperature on the determination of the content of the mulberroside A in the Morus alba (L.) were investigated.On the basis of sigle-factor experiments,the optimum extraction process of mulberroside A in Morus alba (L.) is methanol concentration (60%),solvent volume (55 mL),the extraction time (40 min) and the extraction temperature (35 ℃).Under the optimal extraction condition,the mulberroside A productivity was 7.63 mg/g,which was in consistent with the predicted value of 7.64 mg/g.The calibration curve was in good linearity within the range from 0.0128 to 0.064 mg/mL(r=0.999 3).The content of mulberroside A in Morus alba (L.) in defferent areas was 4.97~13.14 mg/g.The preferred process of optimizing the extraction process of mulberroside A in Morus alba (L.) with response surface analysis is stable and feasible,and can be used for the determination of mulberroside A in Morus alba (L.).
