虎杖主要成分的分离纯化及抗高尿酸血症作用研究

Separation and purification of the main components from Polygonum cuspidatum and their anti-hyperuricemia effect

摘要:

本研究主要探讨虎杖提取物中主要化合物的分离纯化方法,并分别考察其对高尿酸血症大鼠的降尿酸作用。采用大孔树脂法和高速逆流法分离虎杖中三种主要化合物,虎杖苷、白藜芦醇和大黄素。采用氧嗪酸钾对大鼠进行了高尿酸血症造模,测定大鼠血清中尿酸、肌酐和尿酸氮水平。结果发现大孔树脂法可以分离纯化得到虎杖苷和白藜芦醇,高速逆流法可以分离得到大黄素。虎杖苷、白藜芦醇、大黄素可以显著降低高尿酸血症大鼠血清中的尿酸水平。综上所述,虎杖中的三种主要化合物可以通过大孔树脂和高速逆流技术分离纯化,并且都具有显著的降尿酸作用。

Abstract:

The isolation and purification methods of the main components from Polygonum cuspidatum were established and the activity of the main components on anti-hyperuricemia was investigated.The microporous resin combined with high speed current chromatography (HSCCC) were used to separate and purify the three compounds,namely polydatin,resveratrol and emodin.The hyperuricemia of rat model was made by oxonic acid potassium salt.The level of uric acid (UA),creatinine (Cr) and blood urea nitrogen (BUN) in rat serum were measured.The results showed that polydatin and resveratrol were separated by macroporous resin and emodin by HSCCC.The levels of UA,Cr,BUN of the hyperuricemia of rat model were increased comparing to the normal rats.Based on the hyperuricemia of rat model,polydatin,resveratrol and emodin significantly reduced the level of UA.In conclusion,polydatin, resveratrol and emodin from P. cuspidatum were rapidly separated and purified by macroporous resin and HSCCC,and the three main compounds exhibited an obviously positive effect on hyperuricemia rat.
