Biological Transformation of Ardipusilloside I by Human Intestinal Bacteria in vitro

Abstract: Ardipusilloside I (ADS I) and the fresh fecal extract of healthy people were incubated under anaerobic conditions at 37 ℃ for 72 h,and the incubating solution was pretreated by liquidliquid extraction,rotary evaporation and redissolving method sequentially.High performance liquid chromatography-evaporative light scattering detector method (HPLC-ELSD) and ultra-high performance liquid chromatography-electrospray ion-tandem mass spectrometry method (UHPLC-ESI-MS/MS) were used to deduce the chemical structures of ADS I biotransformation products.It was found that deglycosylation reactions occured when ADS I was incubated with human intestinal bacteria.The chemical structures of ADS I biotransformation products was initially deduced,including three secondary glucosides (M1,M2,M3) and one aglycone (M4).The biotransformation of natural product ADS I in human intestinal bacteria in vitro was studied for the first time.It provided the possible scientific evidence for ADS I biotransformation and absorption in human intestinal.
