沙棘中一种黄酮苷在大鼠体内的药代动力学及代谢研究

摘要:

为探究沙棘中一种黄酮苷——山奈酚-3-O-β-D-槐二糖-7-O-α-L-鼠李糖在体内代谢过程,本文采用LC-MS/MS法,建立其在大鼠体内的药代动力学及代谢的检测方法。结果表明,山奈酚-3-O-β-D-槐二糖-7-O-α-L-鼠李糖在体内具有快速清除(T=4~6 h)和较低的相对生物利用度(0.33%±0.09%)的特点。体内分布实验表明,该化合物主要分布在肝脏组织。代谢实验中发现,该化合物主要以原形及脱去7-O-鼠李糖的形式排出体外。本研究初步阐明了山奈酚-3-O-β-D-槐二糖-7-O-α-L-鼠李糖在体内的代谢过程,为探究沙棘中的黄酮苷在体内活性成分提供了理论基础。

Abstract:

Kaempferol-3-3-O-β-D-sophoroside-7-O-α-L-rhamnoside (KSR) is isolated from Hippophae rhamnoides subsp. sinensis.In this study,in order to better study the active constituents in vivo,the ADME (absorption,distribution,metabolism and excretion) of KSR were also studied by LC-MS/MS.In the pharmacokinetic analysis,KSR exhibited high elimination (T=4-6 h) and poor absolute bioavailability (0.33%± 0.09%) after oral administration.In the tissue distribution,the content of KSR was highest in liver tissue.In excretion,KSR was excreted in the form of detracting 7-O-rhamnose from the body.In this study,we initially elucidated the pharmacokinetics and metabolites of KSR,which may have an important significance to study flavonoids in Hippophae rhamnoides L.
